Will there be bacterial growth if both carbenicillin and zeocin are added to the culture medium of pGIPZ?
You should be able to grow pGIPZ constructs in carbenicillin and zeocin together. The recommendation that the clones be grown in carbenicillin only is simply to increase plasmid DNA yield during a plasmid prep because we observed better yields of plasmid DNA if zeocin is not used during culturing for plasmid preps.
Zeocin does not hamper the bacterial growth. If you are having difficulty growing the clones, we suggest spinning the stock culture tubes down to pellet the cells and then take from the pellet to inoculate a starter culture.
