Why did my shRNA arrive as a glycerol stock?
shRNAs can be provided in a vector format: either a plasmid or viral particle. The glycerol stock of bacteria you received is transformed with a plasmid containing the shRNA construct you ordered. You can expand your glycerol stock as described in the product specific technical manual (Example: SMARTvector™ Lentiviral shRNA) , isolate the plasmid contained in this stock, then perform a transfection on your cell line of interest. Alternatively, this plasmid can be packaged into lentiviral particles using a second-generation packaging kit. In this case, your cells will be transduced with the viral construct you generate.
