What are the best positive controls siRNAs to use?
The best positive control is an internal control that is under normal transcriptional regulation. Dharmacon has developed a number of internal siRNA positive controls to target standard, endogenously expressed housekeeping genes such as human and mouse cyclophilin B and lamin A/C. Under the proper transfection conditions these siRNAs will reduce the target signal at the mRNA level by at least 75%. Alternatively, siRNA positive controls directed against reporter genes (e.g., luciferase, GFP) may also be used.
