How are fluorescently labeled siRNAs detected?
Fluorophore labeled duplexes are popular items for optimizing transfection conditions. Uptake of the labeled pre-synthesized siRNAs is readily detected either by flow cytometry or by fluorescence confocal microscopy. We recommend that you use protocols and procedures that are specific for your optical system and software package to detect the fluorescently labeled cells. It is also important to consider that fluorescence may not correlate with siRNA function unless the siRNA has been modified specifically for nuclease resistance (e.g., siGLO control duplexes).
