Edit-R All-in-one Lentiviral sgRNA Positive Controls and kits are designed to target human, mouse and rat genes and can be included in all gene editing experiments using the all-in-one format to confirm successful transduction by observing the generation of insertions and deletions (indels) in your targeted genomic DNA using simple DNA mismatch detection assays.

Individual Edit-R All-in-one Lentiviral sgRNA Positive Controls are available as glycerol stocks and as concentrated, purified lentiviral particles (50 µL at ≥ 1 x 107 TU/mL); available for human, mouse, or rat and designed against:

  • Cyclophilin B (PPIB)
  • DNA (cytosine-5)-methyltransferase 3B (DNMT3B)

Edit-R Lentiviral sgRNA Positive Control Kits include:

  • Lentiviral particles or glycerol stock
  • DNA mismatch detection primers (forward and reverse)
The Edit-R All-in-one Lentiviral sgRNA platform expresses both critical components based on the natural S. pyogenes system in an efficient single vector format; Cas9 and a gene-specific sgRNA  designed to the target site of interest. To facilitate rapid generation of cells lines that constitutively express Cas9 nuclease and the sgRNA, the vectors are available pre-packaged into lentiviral particles that have been purified and concentrated. Cell lines can be easily generated with the Edit-R All-in-one Lentiviral sgRNA particles and gene knockouts can be readily evaluated.

In the Edit-R All-in-one Lentiviral sgRNA vector backbone, the gene-specific guide RNA is expressed under the control of a human U6 promoter, while expression of the Cas9 and puromycin resistance marker (PuroR) is driven from either the human EF1α or the mouse CMV promoter. The plasmid contains the AmpR resistance marker for growth and selection in E. coli.

For the two-vector system, cells were previously transduced with a constitutive hEF1α- or mCMV-Cas9 expression lentiviral particles at an MOI of 0.3, and selected with 10 µg/mL blasticidin for 10 days, followed by transduction with Non-targeting control (NTC), DNMT3B- or PPIB-sgRNA lentiviral particles at an MOI of 0.3. For the all-in-one system, wild-type cells were transduced with the all-in-one were selected with 2 µg/mL puromycin for 3 days. For the all-in-one system, wild-type cells were transduced with the NTC, DNMT3B or PPIB all-in-one lentiviral particles that also expresses a constitutive hEF1α- or mCMV-Cas9 nuclease at an MOI of 0.3. All transduced cells were selected with 2 µg/mL puromycin for 3 days. The cells were then lysed and analyzed for indels using a DNA mismatch detection assay with T7EI.
