Some of the genes in my library are expressed at very high levels and are unlikely to show strong phenotypes with CRISPRa, why did you include them in the gene list?

Genes that are already highly expressed typically show lower activation (or no activation) by CRISPRa. We provide guide RNAs against all protein coding genes. The gene expression level will vary by cell type and has to be determined empirically for your cells of interest. Knowing the basal gene expression level is going to be valuable during the hit identification, prioritization and subsequent hit validation steps.
